Condensin II helps sisters find their identity

چکیده
Telocentrosomes organize telomere gathering Y oshida et al. identify a new job for microtubule-organizing centers (MTOCs)—helping telomeres huddle during meiosis. The most famous MTOC is the centrosome, a permanent structure that serves as a hub for the cell’s microtubules and anchors the spindle during mitosis. But cells also deploy ephemeral MTOCs to perform other tasks. In fi ssion yeast, for instance, part-time MTOCs help position the nucleus during mitosis. Yoshida et al. uncovered a new MTOC function in fi ssion yeast while studying the clustering of telomeres, the caps that protect chromosome ends. During meiosis, telomeres crowd together near the nuclear membrane, a maneuver that enables homologous chromosomes to pair. Previous studies suggested that dynein helps bring telomeres together, and the researchers found that kinesin motors also provide a push. Because these molecular motors stick to microtubules, the team investigated how these fi laments affect telomere movements. They discovered that a -tubulin–containing MTOC, which they dubbed the telocentrosome, formed near the telomeres and that its microtubules helped draw the chromosome caps together near the spindle pole body, the yeast equivalent of the centrosome. In cells lacking one protein necessary for telocentrosome formation, telomeres remained aloof, and homologous chromosomes didn’t pair properly. The researchers think that dynein and kinesin interconnect the microtubules linked to the spindle pole body and the telocentrosome, sliding along these fi bers to help reel in the telomeres. Still unknown is what spurs the telocentrosome to form at a specifi c point in meiosis and what causes it to disassemble. Yoshida, M., et al. 2013. J. An SMC-like protein binds and regulates Caenorhabditis elegans condensins
Structural Maintenance of Chromosomes (SMC) family proteins participate in multisubunit complexes that govern chromosome structure and dynamics. SMC-containing condensin complexes create chromosome topologies essential for mitosis/meiosis, gene expression, recombination, and repair. Many eukaryotes have two condensin complexes (I and II); C. elegans has three (I, II, and the X-chromosome specia...
متن کاملCondensin-Dependent rDNA Decatenation Introduces a Temporal Pattern to Chromosome Segregation
The chromosomal condensin complex gives metaphase chromosomes structural stability. In addition, condensin is required for sister-chromatid resolution during their segregation in anaphase [1-7]. How condensin promotes chromosome resolution is poorly understood. Chromosome segregation during anaphase also fails after inactivation of topoisomerase II (topo II), the enzyme that removes catenation ...
متن کاملCondensin II is anchored by TFIIIC and H3K4me3 in the mammalian genome and supports the expression of active dense gene clusters
Structural maintenance of chromosome complexes, such as cohesin, have been implicated in a wide variety of chromatin-dependent functions such as genome organization, replication, and gene expression. How these complexes find their sites of association and affect local chromosomal processes is not well understood. We report that condensin II, a complex distinct from cohesin, physically interacts...
متن کاملA role of topoisomerase II in linking DNA replication to chromosome condensation
The condensin complex and topoisomerase II (topo II) have different biochemical activities in vitro, and both are required for mitotic chromosome condensation. We have used Xenopus egg extracts to investigate the functional interplay between condensin and topo II in chromosome condensation. When unreplicated chromatin is directly converted into chromosomes with single chromatids, the two protei...
متن کاملKIF4 helps mitotic chromosomes get in shape
KIF4 helps mitotic chromosomes get in shape T hough chromosomes spend most of their time occupying amorphous territories in interphase nu-clei, we typically picture them as the compact X-shaped structures that form during mitosis. Chromosomes adopt this conformation to avoid becoming entangled as they segregate into daughter cells, but how mitotic chromatin condenses into the correct shape is u...
